Os livros contêm muito mais detalhes do que os filmes, e nos filmes podemos interpretar e inferir muito da linguagem corporal e da maneira como as falas são faladas, e nos colocando no lugar de um personagem que sofre de licantropia (eu li muito em cenas dele e Tonks nos filmes posteriores) - Estou me perguntando se, na riqueza das informações apresentadas nos livros, Lupin é aludido a estar deprimido / sofrendo de depressão (mesmo que não seja explicitamente declarado)
Vou abordar isso da maneira que um profissional de saúde mental faria se Lupin entrasse no consultório de um psicoterapeuta e descrevesse sua situação. O terapeuta combinaria o que ele diz com os critérios para o que o ICD-10* é classificado como "transtorno depressivo", que é o que a maioria das pessoas quer dizer quando se refere coloquialmente a "depressão".
É claro que, como Lupin não pode descrever diretamente sua situação, teremos que pagar com citações com as quais temos que trabalhar, seja do Harry Potter livros ou Pottermore. Vou focar em Lupin como adulto, e não como criança.

The downfall of Voldemort, such a source of jubilation to the rest of the wizarding community, marked the beginning of a long stretch of loneliness and unhappiness for Remus.
